We also tested the monoamine oxidase (MAO) inhibitor phenelzine. To evaluate whether the effect of phenelzine on HIV reactivation was due to its anti-LSD1 (KDM1) activity or its antiMAO activity, we tested a series of highly specificMAO inhibitors or LSD-1 (KDM1) inhibitors.Glial cells from five individual JRCSF-HA-infected mice were cultured with or without phenelzine for 2 days, and HIV copies in the culture supernatants measured by qRT-PCR. NOD Cg-Prkcdscid Il2rgtm1Wjl/SzJ (NSG) were transplantation with up to 10^6 human CD34+ HSC, conditioned using sub-lethal radiation doses of 150 Gy (neonates) or 250 Gy (adults), or treated with 35 mg/kg busulfan (adults) (Alpha Aesar, Haverhill, MA). After 4 h (irradiation) or 24 h (busulfan), the mice were injected with 1 × 10^6 CD34+ cells, isolated from human fetal liver. Humanized mice between ages 12 to 20 weeks, engrafted with at least 30% human CD45+ cells in the blood, of which at least 10% were CD3+/CD4+, were infected with JRCSF or JRCSF-HA, using 500 μL intraperitoneal injections containing 5 to 10 × 10^5 infectious units.

3.此模型具有完善的人的免疫系统,HIV感染后,具有艾滋病病人相似的临床症状,所以具有很高的比较医学应用价值。
